Modulatory effect on dyslipidemia and anti-atherosclerotic function of in newly diagnosed type 2 diabetes

摘要: The modulatory effect of on dyslipidemia and the preventive effect on atherosclerosis in newly diagnosed type 2 diabetic patients were studied. A prospective clinical trial was conducted to test the effectiveness of in selected 201 newly diagnosed type 2 diabetic patients. All patients were randomly divided into two groups: 108 with therapy and 93 without therapy. The mean follow-up period was 22 months. The blood glucose and blood pressure of all patients were under control. Serum levels of total cholesterol (TC), triglyceride (TG), and low density lipoprotein cholesterol (LDL-C) were significantly lowered and their decreased percentages were significantly higher in therapy group ( < 0.05). The number of patients with arteria iliaca intima thickening was significantly lower in group of therapy ( = 0.024). With stepwise multivariate logistic regression analysis, the decreased percentage of TG was significantly and independently related with the decreased number of patients with arteria iliaca intima thickening ( = 0.005). The study demonstrates that therapy is effective on modulating dyslipidemia in newly diagnosed type 2 diabetes patients, and may be related with the improvement of early atherosclerosis.

Improved dissolution and anti-inflammatory effect of ibuprofen by solid dispersion

The purpose of this study was to improve the dissolution rate and anti-inflammatory effect of ibuprofen by a solid dispersion (SD) method. Initial screening was developed based on drug solubility in carriers in the liquid state to select a suitable water-soluble carrier system for the preparation of SDs. The dissolution of ibuprofen in urea was higher than in PEG4000 or mannitol. Thus, urea was selected as the carrier for the preparation of SDs. SDs were characterized in terms of dissolution, differential scanning calorimetry (DSC), X-ray diffraction (XRD), scanning electron microscopy (SEM), and Fourier transform infrared (FTIR) spectroscopy. Solid dispersion-based (SDBT) and conventional (CT) tablets were prepared by the wet granulation method. The anti-inflammatory effect of SDBT was evaluated using the mouse ear edema test with xylene. In vitro release results indicated that the ibuprofen dissolution rate was improved by the SD. SD characterization results suggested that ibuprofen partly precipitates in crystalline and amorphous forms after SD preparation and that ibuprofen and urea do not interact. SDBT displayed more significant anti-inflammatory effects than CT. The dissolution rate and anti-inflammatory effect of ibuprofen were significantly enhanced by the ibuprofen-urea SD.

Anti-hyperglycemic effect of the polysaccharide fraction isolated from mactra veneriformis

摘要: Total macromolecule extract was obtained from the soft body of by the coupling techniques of decoction and alcohol precipitation. The extract was deproteinized with an ion exchange column, and resulted in the purifying of the crude polysaccharide fraction. It was found by chemical analysis that the crude polysaccharide part is composed of abundant polysaccharides (>95%) and few proteins (<1%). Furthermore, only one type of monosaccharide, glucose, was detected from its hydrolytes by thin-layer chromatography, indicating that the polysaccharides might be analogs of glucosan. The anti-hyperglycemia effects of the crude polysaccharide part were preliminarily investigated using several pharmacological methods in normal and diabetic mice. Animal experimental results showed that the crude polysaccharide fraction exhibited proper glycemia inhibition activity, and 300 mg/kg-weight dose has the optimal effect among all the studied doses. It is concluded that the crude polysaccharide fraction can be explored as a novel health product that possesses potential as an anti-hyperglycemic agent.

城市规划中抗震减灾的新构想——“强震发生断层”的发现和启示

不是所有的活断层都会发生破坏性地震(5~6级以上),只有其中的极少数在未来数十年、数百年内有可能发生强震,特称之为强震发生断层。这种断层有一定宽度,一般为数米、数十米到数百米或更多。在断层范围内,地震波峰值加速度陡增,烈度可增加2°~3°或更多,抗震设计很困难。断层范围外,烈度随着离震中距离的增大而减弱,抗震按常规设计即可;台湾集集地震的观测数据却提示,0.15g 的抗震设计,竟能抵御 0.5g 的破坏,抗震设计采用的地震动参数,蕴藏着巨大的抗震潜能,这使建筑物抗震达到人员零死亡的目标成为可能。强震发生断层的识别和圈定,从地震构造学的角度解决了地震预报的时、空、强三个要素。城市规划中在采用诸多有关抗震规范时,应考虑强震发生断层的有无及其增震作用,提请有关部门对规范作必要的修改和补充。

CD47 blockade improves the therapeutic effect of osimertinib in non-small cell lung cancer

摘要: The third-generation epidermal growth factor receptor (EGFR) inhibitor osimertinib (OSI) has been approved as the first-line treatment for EGFR-mutant non-small cell lung cancer (NSCLC). This study aims to explore a rational combination strategy for enhancing the OSI efficacy. In this study, OSI induced higher CD47 expression, an important anti-phagocytic immune checkpoint, via the NF-κB pathway in EGFR-mutant NSCLC HCC827 and NCI-H1975 cells. The combination treatment of OSI and the anti-CD47 antibody exhibited dramatically increasing phagocytosis in HCC827 and NCI-H1975 cells, which highly relied on the antibody-dependent cellular phagocytosis effect. Consistently, the enhanced phagocytosis index from combination treatment was reversed in CD47 knockout HCC827 cells. Meanwhile, combining the anti-CD47 antibody significantly augmented the anticancer effect of OSI in HCC827 xenograft mice model. Notably, OSI induced the surface exposure of “eat me” signal calreticulin and reduced the expression of immune-inhibitory receptor PD-L1 in cancer cells, which might contribute to the increased phagocytosis on cancer cells pretreated with OSI. In summary, these findings suggest the multidimensional regulation by OSI and encourage the further exploration of combining anti-CD47 antibody with OSI as a new strategy to enhance the anticancer efficacy in EGFR-mutant NSCLC with CD47 activation induced by OSI.

齐岳山隧道F11高压富水断层带注浆施工技术

摘要:

隧道岩溶、断裂带等高压涌水是隧道修建中比较难以解决的问题,隧道突涌水地质灾害频繁发生,成为影响隧道施工和安全的主要障碍。宜万铁路齐岳山隧道所穿越暗河、溶洞、断层破碎带等不良地质高压突涌水地段,给施工带来很大的困难。目前正在施工的齐岳山隧道F11断层破碎带,水压高、水量大、地质条件复杂,成为唯一制约全线的主要技术难题。以平导F11断层的注浆施工治水防坍为实例,介绍F11高水压断裂带治水防坍新方案、新工艺及效果检查新方法等,为以后类似工程积累经验。

Influence of advanced engineering measures on displacement and stress field of surrounding rock in tunnels crossing active strike-slip faults

摘要: Based on significant improvements in engineering materials, three advanced engineering measures have been proposed—super anchor cables, high-strength concrete anti-fault caverns, and grouting modification using high-strength concrete-to resist fault dislocation in the surrounding rock near tunnels crossing active strike-slip faults. Moreover, single- or multiple-joint advanced engineering measures form the local rock mass-anti-fault (LRAF) method. A numerical method was used to investigate the influence of LRAF methods on the stress and displacement fields of the surrounding rock, and the anti-fault effect was evaluated. Finally, the mechanism of action of the anchor cable was verified using a three-dimensional numerical model. The numerical results indicated that the anchor cable and grouting modification reduced the displacement gradient of the local surrounding rock near the tunnels crossing fault. Furthermore, anchor cable and grouting modifications changed the stress field of the rock mass in the modified area. The tensile stress field of the rock mass in the modified anchor cable area was converted into a compressive stress field. The stress field in the modified grouting area changed from shear stress in the fault slip direction to tensile stress in the axial tunnel direction. The anti-fault cavern resisted the dislocation displacement and reduced the maximum dislocation magnitude, displacement gradient, and shear stress. Among the three advanced engineering measures, the anchor cable was the core of the three advanced engineering measures. An anchor cable, combined with other LRAF measures, can form an artificial safety island at the cross-fault position of the rock mass to protect the tunnel. The research results provide a new supporting idea for the surrounding rock of tunnels crossing active strike-slip faults.

Three-dimensional finite difference analysis of shallow sprayed concrete tunnels crossing a reverse faultor a normal fault: A parametric study

摘要: Urban tunnels crossing faults are always at the risk of severe damages. In this paper, the effects of a reverse and a normal fault movement on a transversely crossing shallow shotcreted tunnel are investigated by 3D finite difference analysis. After verifying the accuracy of the numerical simulation predictions with the centrifuge physical model results, a parametric study is then conducted. That is, the effects of various parameters such as the sprayed concrete thickness, the geo-mechanical properties of soil, the tunnel depth, and the fault plane dip angle are studied on the displacements of the ground surface and the tunnel structure, and on the plastic strains of the soil mass around tunnel. The results of each case of reverse and normal faulting are independently discussed and then compared with each other. It is obtained that deeper tunnels show greater displacements for both types of faulting.

Dihydroartemisinin increased the abundance of by YAP1 depression that sensitizes hepatocellular carcinoma to anti-PD

摘要: The effect of anti-programmed cell death 1 (anti-PD-1) immunotherapy is limited in patients with hepatocellular carcinoma (HCC). Yes-associated protein 1 (YAP1) expression increased in liver tumor cells in early HCC, and Akkermansia muciniphila abundance decreased in the colon. The response to anti-PD-1 treatment is associated with A. muciniphila abundance in many tumors. However, the interaction between A. muciniphila abundance and YAP1 expression remains unclear in HCC. Here, anti-PD-1 treatment decreased A. muciniphila abundance in the colon, but increased YAP1 expression in the tumor cells by mice with liver tumors in situ. Mechanistically, hepatocyte-specific Yap1 knockout (Yap1LKO) maintained bile acid homeostasis in the liver, resulting in an increased abundance of A. muciniphila in the colon. Yap1 knockout enhanced anti-PD-1 efficacy. Therefore, YAP1 inhibition is a potential target for increasing A. muciniphila abundance to promote anti-PD-1 efficacy in liver tumors. Dihydroartemisinin (DHA), acting as YAP1 inhibitor, increased A. muciniphila abundance to sensitize anti-PD-1 therapy. A. muciniphila by gavage increased the number and activation of CD8+ T cells in liver tumor niches during DHA treatment or combination with anti-PD-1. Our findings suggested that the combination anti-PD-1 with DHA is an effective strategy for liver tumor treatment.

Curcumin based combination therapy for anti-breast cancer: from

摘要: While drug resistance appears to be an inevitable problem of an increasing number of anticancer drugs in monotherapy, combination drug therapy has become a prosperous method to reduce the administered total drug dosages as well as overcome the drug resistance of carcinoma cells. Curcumin, considered to possess multi-faceted roles in cancer treatment according to its multiple anti-neoplastic mechanisms as a depressor of chemo-resistance, can significantly facilitate its anti-cancer functions and improve therapeutic effects via combination usage with a variety of other drugs with different reaction mechanisms. To explore this possibility, four anti-cancer chemotherapeutic agents that all possess a certain degree of drug resistance problems, including three tyrosine kinase inhibitors (erlotinib, sunitinib and sorafenib) that are acting on different cell pathways and a typical anticancer drug doxorubicin, were combined with curcumin individually to examine the synergistic anti-tumor effect both and . Results revealed that sunitinib combined with curcumin at the molar ratio of 0.46 yielded the most potent synergistic effect , and was therefore chosen for further animal evaluation. To further enhance the anti-cancer effect, bovine serum albumin (BSA) nanoparticles were utilized as a carrier to deliver the selected drug combination . Preliminary findings confirmed our hypothesis of being able to maintain a similar injected drug ratio for prolonged time periods in tested animals by our approach, thereby maximizing the therapeutic potency yet minimizing the toxicity of these drugs. This work could open up a new avenue on combination drug therapy and realization the clinical utility of such drugs.

A modular cable-driven humanoid arm with anti-parallelogram mechanisms and Bowden cables

摘要: This paper proposes a novel modular cable-driven humanoid arm with anti-parallelogram mechanisms (APMs) and Bowden cables. The lightweight arm realizes the advantage of joint independence and the rational layout of the driving units on the base. First, this paper analyzes the kinematic performance of the APM and uses the rolling motion between two ellipses to approximate a pure-circular-rolling motion. Then, a novel type of one-degree-of-freedom (1-DOF) elbow joint is proposed based on this principle, which is also applied to design the 3-DOF wrist and shoulder joints. Next, Bowden cables are used to connect the joints and their driving units to obtain a modular cable-driven arm with excellent joint independence. After that, both the forward and inverse kinematics of the entire arm are analyzed. Last, a humanoid arm prototype was developed, and the assembly velocity, joint motion performance, joint stiffness, load carrying, typical humanoid arm movements, and repeatability were tested to verify the arm performance.

Basic research on machinery fault diagnostics: Past, present, and future trends

Machinery fault diagnosis has progressed over the past decades with the evolution of machineries in terms of complexity and scale. High-value machineries require condition monitoring and fault diagnosis to guarantee their designed functions and performance throughout their lifetime. Research on machinery Fault diagnostics has grown rapidly in recent years. This paper attempts to summarize and review the recent R&D trends in the basic research field of machinery fault diagnosis in terms of four main aspects: Fault mechanism, sensor technique and signal acquisition, signal processing, and intelligent diagnostics. The review discusses the special contributions of Chinese scholars to machinery fault diagnostics. On the basis of the review of basic theory of machinery fault diagnosis and its practical applications in engineering, the paper concludes with a brief discussion on the future trends and challenges in machinery fault diagnosis.

Machine learning for fault diagnosis of high-speed train traction systems: A review

摘要: High-speed trains (HSTs) have the advantages of comfort, efficiency, and convenience and have gradually become the mainstream means of transportation. As the operating scale of HSTs continues to increase, ensuring their safety and reliability has become more imperative. As the core component of HST, the reliability of the traction system has a substantially influence on the train. During the long-term operation of HSTs, the core components of the traction system will inevitably experience different degrees of performance degradation and cause various failures, thus threatening the running safety of the train. Therefore, performing fault monitoring and diagnosis on the traction system of the HST is necessary. In recent years, machine learning has been widely used in various pattern recognition tasks and has demonstrated an excellent performance in traction system fault diagnosis. Machine learning has made considerably advancements in traction system fault diagnosis; however, a comprehensive systematic review is still lacking in this field. This paper primarily aims to review the research and application of machine learning in the field of traction system fault diagnosis and assumes the future development blueprint. First, the structure and function of the HST traction system are briefly introduced. Then, the research and application of machine learning in traction system fault diagnosis are comprehensively and systematically reviewed. Finally, the challenges for accurate fault diagnosis under actual operating conditions are revealed, and the future research trends of machine learning in traction systems are discussed.

Anti-biofouling strategies for implantable biosensors of continuous glucose monitoring systems

摘要: Continuous glucose monitoring (CGM) systems play an increasingly vital role in the glycemic control of patients with diabetes mellitus. However, the immune responses triggered by the implantation of poorly biocompatible sensors have a significant impact on the accuracy and lifetime of CGM systems. In this review, research efforts over the past few years to mitigate the immune responses by enhancing the anti-biofouling ability of sensors are summarized. This review divided these works into active immune engaging strategy and passive immune escape strategy based on their respective mechanisms. In each strategy, the various biocompatible layers on the biosensor surface, such as drug-releasing membranes, hydrogels, hydrophilic membranes, anti-biofouling membranes based on zwitterionic polymers, and bio-mimicking membranes, are described in detail. This review, therefore, provides researchers working on implantable biosensors for CGM systems with vital information, which is likely to aid in the research and development of novel CGM systems with profound anti-biofouling properties.
